چکیده

Abstract Background Intestinal fibrosis is a common complication of inflammatory bowel disease (IBD) with limited diagnostic modalities to detect and determine the degree fibrosis. Fibroblast activation protein alpha (FAP) both dipeptidyl peptidase endopeptidase properties which virtually absent in healthy tissues has increased expression chronic fibrotic diseases. In this study, we investigated presence FAP intestinal caused by Crohn’s (CD) ulcerative colitis (UC). Methods For transmural surgical resection specimens were retrieved from Tytgat institute IBD biobank located at Amsterdam University Medical Center. patients undergoing an ileocecal for stricturing CD or subtotal colectomy therapy-refractory disease, right-sided hemicolectomy dysplasia non-metastasized colorectal cancer (CRC), included. All had established diagnosis (CD, UC, CRC) previous endoscopy biopsies provided informed consent prior surgery. gene levels determined using immunohistochemistry staining quantitative polymerase chain reaction (qPCR). Mass cytometry was used identify FAP–expressing cells. Results total, 59 ileal 35 colonic samples stained quantified asses (figure 1). A 1.7-fold 1.9-fold increase seen inflamed (mean 22.3, p=0.0069); stenotic 25.5 p=0.0002) ileum compared non-IBD tissue 13.1). Inflamed UC colon 26.8, p = 0.0406) showed 1.4-fold 19.2). expression, 66 42 processed qPCR analysis. 5-fold 6.4-fold found 0.011, p=0.0475) 0.014, p=0.0042) ileum, 0.278, p=0.0195) controls (means: 0.002; 0.165). Conclusion This first study reporting potential as biomarker not only CD, but also UC. Significantly Additionally, CD90+GP38+ cells have been identified most present expressing

Auto-antibodies in Patients with Inflammatory Bowel Disease Unclassified

Background: Inflammatory bowel disease unclassified (IBDU) is considered to be an aberrant immune response with loss of tolerance to many antigens. Objective: This paper tries to address whether there is any value to test for auto-antibodies in such patients. Methods: 60 patients with inflammatory bowel disease unclassified participated in the study. Auto-antibodies to nuclear antigen, intestin...

Probiotic yogurt Affects Pro- and Anti-inflammatory Factors in Patients with Inflammatory Bowel Disease

Inflammatory bowel disease (IBD) is an irregular response of immune system accompanied with different inflammatory manifestations including alterations in cytokines. Probiotics are non-pathogenic organisms with probable effects in various conditions such as inflammation. The present study hypothesized whether oral intake of bifidobacterium and lactobacillus in form of probiotic yogurt may ...

Neutrophil derived human S100A12 (EN-RAGE) is strongly expressed during chronic active inflammatory bowel disease.

BACKGROUND Intestinal inflammation in Crohn's disease (CD) and ulcerative colitis (UC) is characterised by an influx of neutrophils into the intestinal mucosa. S100A12 is a calcium binding protein with proinflammatory properties. It is secreted by activated neutrophils and interacts with the multiligand receptor for advanced glycation end products (RAGE). Promising anti-inflammatory effects of ...

Intestinal Epithelium in Inflammatory Bowel Disease

The intestinal epithelium has a strategic position as a protective physical barrier to luminal microbiota and actively contributes to the mucosal immune system. This barrier is mainly formed by a monolayer of specialized intestinal epithelial cells (IECs) that are crucial in maintaining intestinal homeostasis. Therefore, dysregulation within the epithelial layer can increase intestinal permeabi...
